Examination of the Mutagenetic Effect of the Recombinant Staphylokinase

BACKGROUNDAIM: To investigate if the recombinant staphylokinase has the effect of mutagenesis. MAˉTERIAL AND METHODS: Ames test,chromosomal aberration test in CHL cell and micronucleus test in mouse bone marrow were carried out. RESULTS: The recombinant staphylokinase at0.7～40mg/plate induced neither remarkable nor dose-dependent reverse mutagenecity to four auxotrophic mutants TA 97 ,TA 98 ,TA 100 and TA 102 with or without S 9 mix.Concentrations between1.25and5mg/ml induced neither a dose-dependent nor significant increase in number of structural and numerical chromosomal aberrations with or without S 9 mix.At24h after administration,the freˉquencies of micronucleated polychromatic erythrocytes in bone marrow of mice treated with recombinant staphylokinase between125and500mg/kg did not increase compared with the control mice. CONCLUSION: The recombinant staphylokinase has no effect of mutagenesis.
