Determination of gibberellin abbeymycin and abscisic acid in cucumber by high-performance liquid chromatography

Luhuanggua 9 was used as materials for detection of gibberellin abbeymycin (GA_3) and abscisic acid(ABA) by high-performance liquid chromatography(HPLC).The test materials were grinded in liquid nitrogen and lixiviated in acetonitrile,then pigment was eliminated in chloroform,phenolic was compounded in polyving pyrrolidone(PVP). Phytohormones were extracted in ethyl acetate, and finally PT-C_(18) column were used for further purification. GA_3 and ABA were separated and quantified by μ-Band pak C_(18) reversed HPLC with methanol-water at a flow rate of 0.8 ml/min.Detection was made by a UV detector at 252 nm.The frequency of reclamation was GA_3: 80.12% and ABA:81.33%.
