Optimal Condition for Random Amplify Polymorphous DNA (RAPD) in Chinese Cabbage

Random Amplify Polymorphous DNA(RAPD) is an important technique for molecular markers and study on genetic diversity.In order to establish the optimized RAPD detection system,the Chinese cabbage cultivars 413,96 and their F1 were used.The concentration of DNA,Mg2+,Taq DNA polymerase and dNTPs were optimized for RAPD reaction.The results showed that the optimal conditions were 30 ng(1.5 μl) DNA,3 mmol/L Mg2+,0.01 U/μl Taq DNA polymerase,0.4 mmo/L dNTP and 0.8 μmol/L primer in a 20 μl RAPD-PCR reaction mixture.
