Isolation and Purification of Glucoamylase from Aspergillus niger UV-11 and prelimianry Study Multiple Forms of Glucoamylase

Glueoamylase was isolated and purified from commercial products of As pergillus niger mutant strains UV—11 has been shown to exist in multiple orms, seperable by polyacrylamide gel(7.5％)electrophoresis as nearly ocated bands. After ammonium sulfate fractionation at 60％ saturated gel filtration (Sep hadexG—25)and DEAE—Cellulose DE_52 column chromatography, fou fractions(peak Ⅰ—Ⅳ)from the eluate were obtained.The four fractions have been purified 0.92—1.84 fold with a total recovery of 26.6％. Both protein and carbohydrate of crude enzyme and purified fractionsr were tested in PAGE, date showed that the Glucoamylase from A.niger mutant strains UV—11 was a glycoprotein.
