Determination of caffeine by ATR-infrared spectroscopy

In this thesis,a simple,rapid,solvent-free method has been put forward and established for the quantitative analysis of caffeine.Reference substance suitable for caffeine was precisely weighed,by adding the same volume of potassium ferricyanide as an internal standard substance and KBr used as a diluent and mixing them uniformly.Then a series was prepared.The ATR-FTIR absorption spectrums of the series were measured.The characteristic absorption peak of carbonyl in caffeine and the characteristic cyan-peak in potassium ferricyanide as the peaks for quantitative analysis were selected for the determination.By calculating the ratio of the two peak heights between caffeine and potassium ferricyanide.We concluded that the two peak height ratio showed a good linearity(r=0.995 3) with the concentration of caffeine in the solid mixture at the range of 10~250 mg·g-1.Therefore the concentration of the need checking caffeine was obtained.The average recovery was 99.05%.The experiment result of ATR-FTIR was basically in accord with that of HPLC method.The ATR-FTIR method for the quantitative analysis of caffeine is of high accuracy and reliability.This method could be used in the rapid quantitative analysis for caffeine and others as well.
