Study on Isolation, Purification and Glucopeptide Bond of Glucoprotein form Asparagus Cochinchinensis

The crude glucoprotein, named AG,was extracted with cold water from traditional chinese medicine Asparagus Cochinchinensis,and with acetone precipitation, three fractions, AG-a,AG-b and AG-c,were isolated. Applied to a column of DEAE-Cellulose, AG was devided into AG-w with water elution and AG-c with 0.5N sodium chloride elution. AG-c was homogeneous on gel chromatography and on acetyl fiber film electrophoresis. IR spectrums showed all plucoprotein fractions had O-acetyl groups on their polysaccharide parts. β- Elimination revealed the bond between the polysaccharide and the protein of AG-c was O-type glucopeptide one.
