Effect of Embryo Treatment and Hormone Concentration on Tall Fescue Callus Inducton

He Yangpeng et al (Bureau of Forest of Qujiang Prefecture,Quzhou,Zhejiang 324022)  
In order to establish a tissue culture system of tall fescue, the effect of combination of 2,4-D, BA and different ways of seeds treatment on callus induction was studied. Mature tall fescue seeds were used as explants. The results indicated that:①the effect of hormones on callus induction of tall fescue was significant, high concentration of 2,4-D and BA had negative effect on callus induction. ②the different ways of seeds treatment had significant effect on callus induction, the mean rate of callus induction was 11.07% with chopped seeds treatment, however, it was 2.65% without chopped seeds treatment.③the highest rate of callus induction was obtained on MS medium containing 3mg/L 2,4-D. ④BA had negative effect on callus induction, the higher the concentration was, the more intensive the inhibition was.⑤the best induction medium was MS+3?mg/L 2,4-D+0?mg/L BA.
