Isolation of a Phytase-producing Aspergillus.sp Strain and Optimization of its Fermentation Conditions

A strain X19 capable of producing high temperatur phytase was isolated and identified as Aspergillus sp.After mutated by UV and TNG,a strain can produce higher temperatur phytase was get and named as A73 whose remain activity was 93 percent under 90 ℃ for 10 minutes.Some factors affecting the phytase production from A73 were studied.The results showed that the optimal medium compositions for phytase production were 1.0% glucose,6.0% corn starch,1.0% NaNO_3,0.005% KH_2PO_4,0.05% MgSO_4·7H_2O,0.05% KCl,0.001% MnSO_4·4H_2O and 0.001% FeSO_4·7H_2O.The optimal culture condition is initial pH 5.5,tenperature 28?℃,rotation 200?r/min and fermentation time 96 h.As a result,the phytase activity could reach 2 186 U/ml.
