Effect of Metal Ion on the Polysaccharide Synthesis in Protocorm-like Body of Dendrobium huoshanense in Suspension Culture

This research is concerned with the effect of Mg~(2+),Ca~(2+),Fe~(2+),Mn~(2+)and Zn~(2+) on polysaccharide synthesis in Protocorm-like body of Dendrobium huoshanense in suspension culture through the orthogonal matrix method with one factor-at-a-time.The results showed that the optimal combination for polysaccharide accumulation in protocorm-like body was 1.5 mmol/L Mg~(2+),4.5 mmol/ L Ca~(2+),0.1 mmol/L Fe~(2+),0.5 mmol/L Mn~(2+) and 0.06 mmol/L Zn~(2+).The peak of the polysaccharide production(5.87 g/L) was obtained in the culture medium with the same ion combination mentioned above,which was increased 2.49-fold than that of the former experiment and was increased 31-fold than that obtained in standard MS.Furthermore,the responsible order of metal ion in the polysaccharide synthesis was Mg~(2+) Ca~(2+) Mn~(2+) Fe~(2+) Zn~(2+).
