Effect of Sucrose on Growth of Protocorms of Dendrobium candidum and Accumulation of Polysaccharide

On the basis of the determination of the suitable carbon source,the biomass and the accumulation of polysaccharide from protocorms of Dendrobium candium were studied under different sucrose concentration.The results showed that sucrose and glucose were better than fructose and lactose for protocorms growth and polysaccharide accumulation at the concentration of 30~60 g/L,the inhibitory effect on the growth of protocorms increased,and the growth cycle was becoming longer with sucrose concentration increasing.But the content of polysaccharide increased.Moreover,the sucrose presented inhibition effect on protocorms growth while its concentration was above 60 g/L.The optimum concentration of sucrose for accumulation of polysaccharides was 30 g/L.The maximum biomass of protocorms was 17.93 g/L.The content of polysaccharide reached 208.7 mg/g.And the yield of polysaccharide was 3 679.6 mg/L after 40 day's incubation.
