Optimization of Extraction Technique of Opuntia Polysaccharide via Response Surface Methodology

Effects of different temperature,time and the ratio of solid to solution on the extraction yield of polysaccharide from Opuntia were investigated based on composite design of quadratic regression.Extraction technique parameters were optimized with Response Surface Methodology.Experimental data were analyzed by solving the regression equation with MATLAB 7.0 software.It was indicated that the optimum extraction parameters were 74 ℃,132 min and 276ml water volume.Under those conditions,the predicted value of polysaccharide extraction yield from Opuntia was 89.77 μg/ml,which was in consistent with the measured value of 89.48 μg/ml.This method with optimized technique parameters had maximal extraction yield of polysaccharide from Opuntia.
