Optimization of the Operation Rules for SSR Molecular Marker Technique in Maize

[Objective] The research aimed to establish the operation rules for SSR marker technique in maize and provide basis for its application in the production.[Method] With 2 maize hybrids(Qiandan 16 and Xishan 99) and 4 maize inbred lines(Su 37,Jiao 51,Huang C and 4011) as tested materials,the genomic DNA was extracted from maize.And different DNA extraction methods,SSR reaction systems,PCR amplified procedures and silver staining methods were compared resp.[Result] The amplification effects of the DNA extracted by using SDS method or CTAB method were better.The optimum SSR reaction system in maize was as follows: 1U Taq enzyme,1×Bufer,60 ng template DNA,2.0 mmol/L Mg2+,0.15 mmol/L dNTPs,0.3 μmol/L primer.The optimum SSR amplification procedure was as follows: 93 ℃ for 1 min;30 cycles of 93 ℃ for 1 min,60 ℃ for 2 min,and 72 ℃ for 2 min;72 ℃ for 5 min.It was better to detect maize SSR marker by using 8% non-denaturing polyacrylamide gel electrophoresis or 6% denaturing polyacrylamide gel.[Conclusion] The genetic diversity of 51 maize hybrids in Guizhou were analyzed by using the optimized SSR marker detection system and most primers amplified clear bands.
