Identification of Seed Purity in F1 Pepper Hybrid Cultivar Guangjiao 6 by RAPD

[Objective] The research aimed to lay the foundation for detecting the seed purify of crops by using RAPD technology.[Method] The seed purity of F1 pepper hybrid cultivar Guangjiao 6 was detected by RAPD technology to study the influences of the dosage of template DNA and pnductions in RAPD reaction system on the effects of PCR amplification.[Result] 10～400 ng template DNA was added to 25 μl reaction system and accordant primers could all be obtained with no obvious difference of band brightness.15 difference primers with good stability were screened out from 340 RAPD random primers,among which there was 5 partial-female types of primers,6 partial-male types and 4 complementary types.Three screened polymorphic primers F05,F15 and I05 were used to detect the purity of Guangjiao 6 and the detecting results of 3 primers were same and accordant with the results of morphological identification in the field.[Conclusion] RAPD technique was an effective method of identifying the purity of F1 pepper hybrid cultivar,with the characteristics of exact,reliable,rapid and economic.
