Study on the Karyotype of Peanut Variety Haihua 1

[Objective] The study aimed to analyze karyotype of peanut variety Haihua 1 and accumulate the cytological data of peanut germplasm resources.[Method] With Haihua 1 as tested material,after its root tip,1~2 cm length,was pretreated,fixed and hydrolyzed with enzyme,cell suspension was extracted to make the object plate.Then object plates with good dispersion and clear karyotype were collected and taken photo for karyotype analysis by Adobe Photoshop7.0.[Result] Haihua 1 had the chromosome numbers of 2n=40 and had a pair of satellite chromosome in 15th pair of chromosome.Karyotype formula of Haihua 1 was K(2n)=40=36 m+2 m(SAT)+2Sm.The relative length of chromosome relative length of Haihua 1 ranged from 2.22 to 3.57 and its difference value was 1.35.The total length of chromosome was 96.16 μm and As.K % of Haihua 1 was 60.44%.[Conclusion] Haihua 1 was diploid(2n=40),and its chromosomes were all long,with dissymmetry to some degree.
