Study on the Separation and Identification of Polyphenol in Lotus Root by Thin-layer Chromatography

[Objective] The research aimed to lay the foundation for separating and identifying polyphenol from lotus root.[Method] With silica gel G60 as stationary phase,with toluene-ethyl acetate-icy acetic acid as mobile phase,with catechin as control substance,with FeCl3-K3Fe(CN)6 as chromogenic reagent,phenolic compounds were separated and identified from ester-phase components of polyphenol in lotus root by using thin-layer chromatography(TLC).And the effects of different developing solvent on the separation of components were analyzed.[Result] Rf values between the 2nd spot evolved by ester-phase of polyphenol in lotus root and the spot of standard reagent catechin was the same,which preliminarily confirmed that there was catechin in the ester-phase of polyphenol in lotus root.Better separation effects all could be obtained by using 6 kinds of developing reagents in TLC and their Rf was in the range of 0.49～0.61,which accorded the qualitative demands of Rf in TLC.[Conclusion] TLC was highly efficient,sensitive and rapid,which provided better detection method for preliminarily identifying the compounds of catechin in lotus root.
