mtDNA-RAPD Analysis on Hybrid Second Generation of 99B×island Cotton

[Objective] The aim was to provide reference for the feasibility of detecting the purity of hybrid progenies by mtDNA-RAPD technology and provide basis for the wide application of primer combination method in the plant purity detection and genetic diversity analysis.[Method] mt DNA of 99B,island cotton and their hybrid progeny F2 was amplified by RAPD-PCR using 6 random single-primers and primer combination and then analyzed the polymorphism.[Result] The polymorphism analysis on the mtDNA of hybrid progeny F2 through RAPD detection by using single-primer showed that 3 different strains were obtained from 6 single primers after RAPD-PCR amplification and the purity of the population was 95%.The polymorphism analysis using primer combination showed that 4 different strains were detected from 15 pairs of primers after RAPD-PCR amplification and the purity of the population was 93%.The above result showed that the polymorphism analysis after amplification using primer combination had high accuracy and good veracity and it also showed that primer combination method had advantages and utilization value in the purity detection of plant germplasm.[Conclusion] The polymorphism detection had good utilization value and application prospect.
