Effects of Extraction Methods on Anthraquinone Components and the Antioxidant Activity in Rhubarb

[Objective ] The research aimed to compare the effects of different extraction methods on anthraquinone components and antioxidant activity in rhubarb. [Method] Free anthraquinone and total anthraquinone were extracted by 4 kinds of extraction methods including ultrasonic method,reflux method,Soxhlet method and microwave method and their contents were determined.Their antioxidant activities were determined by the methods of DPPH and FRAP.And the correlation between anthraquinone components and the antioxidant activity was analyzed.[Result] The content of free anthraquinone by ultrasonic extraction method was the highest(0.949%)with significant difference with that by other extraction methods(P0.05).The content of total anthraquinone by ultrasonic extraction method was the highest(1.017%) with significant difference with microwave method and reflux method(P0.05).The determination results of the antioxidant activity showed that the antioxidant ability and the scavenging ability by reflux method was the strongest with FRAP and DPPH of-0.956 3 and-0.952 3 respectively.They had significant difference with that by ultrasonic method and Soxhlet method(P0.05).The correlation analysis showed that total anthraquinone content had negative correlation with FRAP and DPPH,with the correlation coefficients of-0.956 3 and-0.952 3 respectively.[Conclusion] Different extraction methods affected anthraquinone components and the antioxidant activity.Total anthraquinone content had negative correlation with the antioxidant activity.
