Determination of Contents of ATP,ADP and AMP in Medial Gastrocnemius and Soleus Muscle of Rat by HPLC

DAI Zhong-he et al(School of Life Science,Nantong University,Nantong,Jiangsu 226007)
[Objective] The ATP,ADP and AMP contents of soleus muscle and medial gastrocnemius were compared in adult S.D.rats.[Method] The soleus muscle and medial gastrocnemius were obtained and pulverized respectively to muscular cell syrup together with liquid nitrogen and specimen buffer solution such as joining EDTA,HClO4 and NaHCO3 early or late. After high speed freezing, the pure liquid was taken out and the ATP,ADP and AMP contents was measured one by one with HPLC.[Result] The ADP content of soleus muscle was more than that of medial gastrocnemius. The soleus muscle was suited to aerobic movement. The ATP and AMP contents of soleus muscle were less than those of fast muscle-medial gastrocnemius.[Conclusion] The difference of fast and slow muscle could be distinguished with HPLC.
