Study on Ultrasonic Wave Assisted Extraction of Polysaccharide from Morchella esculenta Mycelium

[Objective] The aim was to study the condition for ultrasonic wave assisted extraction of polysaccharide from Morchella esculenta.[Method] With the water as solvent,the polysaccharide from M.esculenta mycelium through fermentation culture was obtained by ultrasonic wave assisted extraction.The influences of extraction time,extraction temperature,extraction times,ultrasonic wave power and solid-liquid ratio on polysaccharide extraction rate were studied.[Result] The polysaccharide extraction rate was highest when the ultrasonic wave power was 400 W.The polysaccharide extraction rate was first increased and then decreased with the prolonging of the extraction time and increase of the solid-liquid ratio.The optimum condition for ultrasonic wave assisted extraction of polysaccharide from M.esculenta was solid-liquid ratio of 1∶10(g/ml),ultrasonic wave power of 400 W,extraction time of 0.75 h,extraction temperature of 55 ℃,extraction times of 2.Under the condition,the extraction rate of polysaccharide from M.esculenta was 0.786 2% and the purity of obtained polysaccharide was higher,the polysaccharide content was 72.81% and the protein content in polysaccharide was 0.92%.[Conclusion] Ultrasonic wave assisted extraction of polysaccharide from M.esculenta had some advantages such as short time,saving energy sources and higher extraction rate.
