Research on the Establishment of Molecular Marker System for Anti-smut Wire of Sweet Sorghum

[Objective] The research progress of the molecular marker of sweet sorghum smut wire gene was reviewed.[Method] The sweet sorghum variety with antismut silk——TR102 and the sweet sorghum variety with the sensitive to head smut——LTR106 being used as experimental material,the SSR technology was preliminarily applied in the analysis of sweet sorghum smut silk gene.The sweet sorghum genome can be extracted with the method of CTAB.[Results] The concentration of Taq polymerase was further optimized through the comparison test of the different concentrations of Taq polymerase.The amplified polymorphism of SSR molecular marker was done by 20 SSR primers and 16 primers were able to clearly amplify the polymorphic bands,which can be used in the initial loci of the resistant gene.Among them,the different band was amplified in 4 primers,which were Xtxp 279,Xtxp284,Xtxp36 and Xtxp228,respectively.[Conclusion] The optimization of the SSR reaction system was realized based on the comparison of different factors.
