Study on Tissue Culture and Preventing Vitrification of Thymus quinquecostatus

[Objective] The study was to provide the basis for the development and utilization of Thymus quinquecostatus.[Method] With the stem with axillary buds as material,after disinfection,they were inoculated on the MS medium.The effects of different culture conditions on subculture multiplication,preventing vitrification,rooting and transplanting effect were investigated.[Result] The optimum treatment for the disinfection on T.quinquecostatus explants was that it was disinfected by 70% ethanol for 45 s,then treated by 0.1% mercuric chloride for 8 min.With the increase of 6-BA concn.,the multiplication coefficient and vitrification of T.quinquecostatus risen significantly.The basal medium MS added by 0.5 mg/L 6-BA and 0.1 mg/L IAA was more optimal for the multiplication culture of T.quinquecostatus.The concn.of 6-BA had an obvious influence on the subculture multiplication culture of T.quinquecostatus.When 6-BA was 0.1 mg/L,sucrose was not lower than 45 g/L and T.quinquecostatus was cultured with different ventilative closure,the vitrification phenomenon was thoroughly eliminated.The rooting rate of medium with 0.2 mg/L IAA was significantly higher than that medium without IAA.After transplanted for 45 d,the survival rate of T.quinquecostatus tissue seedling reached 81.2%.[Conclusion] The optimum culture condition for tissue seedling was obtained.
