Study on Extraction of Actinidia Genomic DNA

[Objective]The purpose of research aimed to discuss a method of extracting the total genomic DNA from Actinidia.[Method]With Actinidia which is abundant in secondary metabolites of polysaccharide and polyphenol as materials,the extraction effect of different variety and method on the genomic DNA of Actinidia was compared with,and influence of the different part of tissue culture,antioxidant and the method of DNA purification on the yield of genomic DNA was studied.[Result]The results indicated that the high quantity of genomic DNA was acquired when the stem of tissue culture plants was used as the material,and CTAB method was used,adding 1% β-Mercaptoethanol into the extraction buffer,and 1/2 volume of 5 mol/L NaCl was added before the precipitation of isopropyl alcohol.[Conclusion]This study will establish basis for molecule biology and gene engineering of Actinidia in future research.
