Observation on Cytological Structure of Two Types of Callus of Glycyrrhiza uralensis Fisch by Microscope

[Objective] The study aimed to compare the difference on the cytological microstructure between the callus from the stem segment and the callus of root segment from Glycyrrhiza uralensis Fisch.[Method] With the callus with redifferentiation ability from the bud in stem segment and the callus without redifferentiation ability from root segment of G.uralensis as the tested materials,they were cultured for 24 d with nonbiological and biological slicing techniques and then their mitochondria and cytoskeletons stained with the living were observed with the smear method and the cytoskeletons were also observed with the paraffin method.[Result] The observation found that the motile mitochondriaes were all in 2 types of callus and there were some significant difference on the number of the motile mitochondriaes in 2 different types of callus cells.The number of the motile mitochondriaes with motor activity in bud callus cells was significantly greater than that in root callus cells.The reticular formations of cytoskeletons in the cells of bud callus were more dense than that in root callus cells.This showed that the cells of bud-induced callus had strong physiological activities and the enough energy that could start the differentiation,which could be easy to make the redifferentiation.The status of the cells of radicle-induced callus was in the opposition.[Conclusion] The difference on the dedifferentiation structure between 2 types of callus had some effects on their redifferentiation.
