Research on the Adsorption and Separation Performance of Macroporous Resin on Total Flavonoid from Sophora japonica

[Objective] The aim was to screen the macroporous adsorption resin with better adsorption and desorption ability on total flavonoid from Sophora japonica and ascertian its optimum adsorption and desorption condition.[Method] The influencing factors of adsorption and desorption performance of resin,such as flow velocity,temperature and pH were reviewed to ascertian its optimum adsorption and desorption condition.[Result] D4020 type nonpolarity macroporous adsorption resin had better adsorption and desorption effect on the total flavonoid from S.japonica.The optimum adsorption condition was pH of 4.5,temperature of 15 ℃,sample concn.of 0.55-0.85 mg/ml and flow velocity of 2.0 BV/h.The desorption condition was 70% ethanol,pH value of 8.5 and flow velocity of 3.0 BV/h.Desorbing resin adsorption D4020 under this condition,3.0 BV desorption solution could desorb more than 90% total flavonoid adsorbed on the resin,then the concn.of desorbed total flavonoid was higher and the eluent could be reclaimed through distillation,which could reduce the cost.[Conclusion] The research provided scientific basis for the adsorption and desorption of total flavonoid from S.japonica.
