Study on the Isolation,Screening of Cellulase-producing Strain and Its Enzymatic Characteristics

[Objective] The research aimed to search out cellulose-producing strain with high yield.[Method] Cellulase-producing strains were screened through primary screening of solid culture and secondary screening of shaking-flask fermentation.Strain B5 with higher cellulase activity were preliminarily studied through main influence factors of B5 such as culture medium,growth temperature,pH value and cellulase-producing time and enzymatic characteristics of the crude enzyme from B5.[Result] Strain B5 with higher cellulase activity and higher stability was screened out.The research results of B5 strain showed that the optimal growth medium for B5 was Charls medium,the optimal growth temperature were 35 ℃,the optimal pH value were 7.5,cellulose-producing time were 72 h.Enzymatic characteristics of B5 showed that the optimal temperature was 50 ℃,the optimal pH value was 7.5.The cellulase of B5 showed a high stability at pH of 7.08.0.[Conclusion] The growth and enzymatic characteristics of B5 were preliminarily confirmed,which provided scientific foundation for further study.
