Study on the Optimization of Extracting Technology of Chlorogenic Acid from Eucommiae Leaf Powder

[Objective] The method and technology of the chlorogenic acid extracted from eucommiae powder were optimized for the reference for the development of the industry.[Method] The optimal technology was experimented with the combination of ultrasound enzymolysis method through the orthogonal test of 4 factors at 3 levels based on the experiment in single factor.[Results] The result from the experiment in the single factor was the maximum production of chlorogenic acid extracted from eucommiae powder was under the following conditions: the pH value of solvent,4.5;the enzyme amount,0.6%;the enzymolysis temperature,40 ℃;the ratio of ethanol to folium eucommiae,1∶10 and the extraction time with ultrasound,30 minutes.The result from orthogonal test indicated that under the optimal extraction technology including the pH value of solvent,5;the enzymolysis temperature,40 ℃;the ratio of ethanol to folium eucommiae,1∶10 and the extraction time with ultrasound,30 minutes.The output of chlorogenic acid was as high as 3.05%.Compared the method of ultrasound and enzymolysis,the output of chlorogenic acid extracted from eucommiae powder was 5.54% higher than that with the enzymolysis method and 16.68% higher than that with the ultrasound method.[Conclusion] The combination of the ultrasound and enzymolysis method was beneficial to the release of chlorogenic acid because the eucommiae powder was firstly reacted by enzymolysis and then by the ultrasound so that the combination method was much better than single way.
