Study on the 12 Pholiota adiposa Strains with the Esterase Isoenzyme

[Objective] The effect of esterase isoezyme on the classification and identification of Pholiota adipose was studied.[Method] With the vertical flat electrophoresis of polyacrylamide gel,12 Pholiota adipose strains from different regions were compared and analyzed in esterase isozyme character and the genetic differences among strains were analyzed with the cluster analysis software.[Results] There were 23 bands detected with electrophoresis staining,the majority of which showed the difference in isozyme patterns.The number of bands among strains was different with from 3 to 14 bands,in which,the band with the value of relative mobility(Rf) at 0.70 was shared by all of 12 strains and at 0.66,by 11 strains,which could be considered as common band.There was very large difference in bands among strain Pa1,Pa2,Pa3,Pa4,Pa5,Pa10 and Pa11 with highly polymorphic character.The strain Pa6 and Pa7 showed basically same band with very close relationship and common genetic basis,which could be different name of same species.The cluster analysis results indicated that the relation degree among 12 strains was from 0.49 and 1.00 and the 12 strains could be divided into 3 species at the relation degree of 0.64.[Conclusion] The esterase isozymes could be used as one of the effective biochemical indicators in the study on the Pholiota adiposa germplasm resources.
