RAPD Analysis of Ornamental Plant "Haierlian" and Its Closely-related Plants

[Objective] The research aimed to study genetic diversity and relationship in Illicium laceolatum of Illicium L..[Method] 13 plant types of Haierlian were used as test materials to make RAPD analysis by using Manglietia fordiana,Magnolia denudate and Cinnamomum camphora as outgroups.[Result] 20 primers with high polymorphism,clear bands and good repeatability were selected from 35 random primers.154 DNA bands were amplified from 13 species of test plants by using selected 20 primers including 47 common bands.The results were analyzed by NTSYSpc 2.10e software and similarity index was 66.2%-90.3%.Using UPGMA method,13 species of tested plants could be clustered into 3 groups.[Conclusion] Haierlian and other tested types have close genetic relationship,whose genetic differences don’t have necessary association with geographic factors.
