Optimization of the Purification of Total Flavonoids from Aurantii immaturus Fructus by Macroporous Resin

[Objective]To investigate the purification conditions of total flavonoids from Aurantii immaturus Fructus by using macroporous resin.[Method] The adsorption quantity and desorption rate of three kinds of macroporous resins(AB-8,HPD-450,D101)were compared,and the seperation parameters of D101 on total flavonoids were studied.[Result] D101 most fitted for the separation of total flavonoids from Aurantii immaturus Fructus.The best adsorption conditions are 3 mg/ml of tatal flavonoids concentration,sample volume is 2 times of the bed volume(BV),at pH value 4.5.Whereas the optimal desorption condition was to elute with 70% ethanol and collect the ethanol solution at 2.0 BV volume.[Conclusion]The D101 macroreticular resin showed better comprehensive adsorption property and can be used to separate and purify the total flavonoids from Aurantii immaturus Fructus.
