Caffeine Content Determination of Bowl-shaped Brick Tea in Different Years by HPLC

[Objective]The aim was to compare the caffeine contents of bowl-shaped brick tea in different years by HPLC.[Method]The water-boiling method was adopted to extract the caffeine from the bowl-shaped brick tea in different years,the cenlrifeged extracts was separated by C18 column and then detected by HPLC-UV.A gradient elution with the methanol solution as the mobile phase A and 0.4% phosphoric solution as mobile phase B was conducted at a flow rate of 1.0 ml/min,and the detection wavelength was 274 nm,the column temperature was 25 ℃,the added quantity was 10 ul.[Results]Good linearity was seen of caffeine in the range of 0.218-2.180 μg,the regression equation Y=315.779X+25.956,r=0.999 9.The average recovery rate of caffeine was 101.3%,RSD was 0.36%.The detected caffeine content in the five years of bowl-shaped brick tea were 19.307,20.114,20.270,19.739,19.733 mg/g respectively with an average of 19.832 mg/g,RSD was 1.90%.[Conclusion]The research results showed that the caffeine content in bowl-shaped brick tea changed small with the growth time.
