Determination of Chlorogenic Acid of Different Organs from Taraxacum mongolicum by High Performance Liquid Chromatography

[Objective] To establish a method for determining contents of chlorogenic acid in different organs from Taraxacum mongolicum by HPLC.[Method] The HPLC was carried out with Shim-pack VP-ODS column(4.6 mm×150.0 mm,5 μm) using mixture of methanol and 0.4 % phosphoric acid solution in different ratio as mobile phase.Other conditions were: flow rate of 1.0 ml/min,detection wavelength of 326 nm,column temperature of 40 ℃ and sampling amount of 20 μl.[Result] When the concentration of chlorogenic acid ranged from 0.791 2 to 79.120 0 μg/ml,the linear relationship between concentration and peak area was good(r=0.999 8).The recovery was 98.00%-101.50%.Leaves、roots、flowers and scapes of Taraxacum mongolicum were 1.212,0.536,0.448 and 0.417 mg/g in chlorogenic acid,respectively.[Conclusion] The method established in this work was a rapid and accurate method for determining chlorogenic acid from Taraxacum mongolicum.Leaf of Taraxacum mongolicum was highest in chlorogenic acid,which provided basis for better developing and applying of Taraxacum mongolicum.
