Determination of rutin in the leaf of hibiscus mutabilisL. by HPLC

Aim To establish a method for the determination of rutin in the leaf of hibiscus mutabilisL.Methods The chromatographic system consisted of ODS column and mobile phase of methyl alcohol-water-acetic acid(30∶70∶2.5),and the detective wavelength was at 254 nm.Results The method was precise with RSD of 1.40% for within-day variation(n=5) and of 0.85% reproducibility(n=5).Average recovery for chlorogeric acid was 103.57% with RSD of 1.27%(n=6).3 samples were analyzed and the contents of chlorogeric acid were from 0.335% to 0.446%.Conclusion This method is proved to be simple and reliable and can be used for the determination of rutin in the leaf of Hibiscus mutabilisL.
