Experimental Study on Curcumin in the Prevention of Renal Interstitial Fibrosis in Rats

Objective:To study the effect of Curcumin on the expression of transforming growth factor-β1(TGF-β1)and connective tissue growth factor(CTGF) on the process of renal interstitial fibrosis caused by unilateral ureteral obstruction(UUO) in rats.Methods:Fifty-four Sprague-Dawley rats were randomly divided into three groups: Sham operated group,UUO model group,Curcumin-treated group.Each group has eighteen rats.The rats in the Curcumin-treated group and UUO model group underwent UUO,rats in sham group had their ureters manipulated but not ligated.Six rats of each group were killed respectively at 3,7,14 days after surgery.The level of BUN and SCr were observed.Histological changes were observed by hematoxylin eosin and Masson staining. Immunohistochemistry was also performed on renal tissue for TGF-β1 and CTGF.Results:In the obstructed kidneys,there were significant renal interstieial fibrosis and interstitial space expansion.Immunohistochemical staining showed that the expression of TGFβ-1 and CTGF significantly increased in the obstructed kidneys.Curcumin could obviously decrease interstitial volume and the expression of TGF-β1 and CTGF,It also improved the histological changes of the UUO renal tissue.Conclusions: Curcumin can down-regulate the expression of TGF-β1 and CTGF in UUO rats,showing its potential ability to inhibit renal interstitial fibrosis caused by UUO in rats.
