Gypenosides promotes the programmed cell death of human liver carcinoma cells

Objecti:To investigate the effect and the mechanism of gypenosides(GPS) on the programmed cell death(PCD) of human liver carcinoma cell line HepG 2-A 16 . Methods:The MTT colorimetric assay and the agarose cultured colony formation micro method were used to investigate the inhibition effect of GPS on the proliferation and colony formation ability in vitro cultured human liver carcinoma(HepG 2-A 16 )cells.DNA fragment ation method and TUNEL method were used to observe the effect of GPS on PCD of HepG 2-A 16 cells .The influence of GPS on Bcl-2 protein expression of HepG 2-A 16 cells was detected by Streptavidin-peroxidase(S-P)immunohistochemical method. Results:1.GPS(50-250 mg/L)inhibted the proliferation and colony formation ability of HepG 2-A 16 cells in a concentration-dependent manner( P 0.05-0.001).2.HepG 2-A 16 cells treated with GPS(50-250 mg/L)for 24 hours showed DNA fragment ladder and bluepurplish grain which are characteristic appearance of PCD by DNA agarose gel electrophoresis and TUNEL staining.3.Bcl-2 protein stain of HepG 2-A 16 cells is positive by S-P immunohistochemical method, whereas it is negtive in those treated with GPS(50 mg/L) for 24 hours. Conclusions:GPS can promote PCD of human liver carcinoma cell and down-regulation of bcl-2 protein expression may be one of the mechanisms which GPS promote PCD of human liver carcinoma cells.
