Expression of cell-cycle regulators cyclin D1 and CDK4 in esophageal squamous cell carcinoma

Objective: To Obtain further information about the role of some faetors regulating the G1 cell cycle in the development of esophageal squamous cell carcinoma (ESC). Methods:A total of ten normal esophageal epithelia and 50 ESC samples were stained immunohi stochemic ally us lug monoclonal antibodies against cyclin D1 and CDK4. Results: cyclin D1 and CDK4 showed low expression in normal esophageal epithelium while overexpression in ESC. Twenty-seven out of fifty cases of ESC showed positive reactivity in cyclin D1 staining, 8 of them were extensive positive, 12 moderate positive and 7 weak positive. Sixteen out of 27 cyclin D1 positive ESC samples were from patients with lymphomatic metastasis. CDK4 positive cells were found in 26/50 ESC. Twelve of CDK4 positive ESC showed cyclin D1 negative. In contrast, 11 of cyclin DI ESC positive showed CDK4 negative. Fourteen out of fifty ESC showed both cyclin D1 and CDK4 positive whereas 13/ 50 ESC showed no reactivity with cyclin D1 and CDK4. We also found cyclin D1 and CDK4 positive cells in tissues near to ESC. Conclusions: Overexpression of cyclin D1 and CDK4 may involve in the onco-pathogenesis of ESC. The increase expression of cyclin D1 and CDK4 possibly related to the differentiation of ESC. The deregulation of cyclin D1 may be one of the early events during the development of ESC. The close relationship between cyclin D1 overexpression and metastasis indicates that cyclin D1 might provide some cell subtype with activated inclination of metastasis. The fact that negative expresion of both cyclin D1 and CDK4 in the same ESC sample suggests that there might be other mechanism regulating ESC progression.
