The effect of a diterpenoid isolated from pteris semipinnata on the HL-60 cell cycle and its enhanced in vitro cytotoxicity by anticancer drugs

Objective: To observe the effect of 6F, one of the diterpenoids isolated from Pteris semipinnata L. (PsL), on the HL-60 cell cycle kinetics and its enhanced cytotoxicity when combined with other antitumor agents. Methods: Cell cycle was determined using F1ow Cytometry (FCM) analysis, Cytotoxicity assay was tested by MTT method. Results: The FCM data showed that the distribution of HL-60 cells among phases of the cell cycle was changed when treated with 6F. After exposure of the cells to various concentration of 6F for 6h, the S and G2/M phase ratio was increased significantly in a dose-dependent manner. After 24h of exposure, S hpase ratio increased further. The inhibitory rates of 6F combined with 2-CldAdo or CDDP or VCR or 5FU were higher than that of these drugs used alone, q values were more than 0.85. Their enhancement times by 6F were 1.58, 1.53, 1.55, 1.38, respectively. Conclusion: The results indicated that 6F could arrest HL-60 cells in S and G2/M phase, 6F could enhanced the cytotoxicity of the drugs mentioned above. it is known that 2-CldAdo arrests cells in S phase, CDDP and VCR arrest cells in G2/M phase, 5FU arrests cells in G1 phase. Thr different effect of these agents on the various phases of cell cycle kinetics may be partially explained the enhanced effect of 6F.
