The effect of retinoic acid on growth inhibition and p53、c mycexpression in gastric carcinoma cells

Objective: To evaluate the effect of retinoic acid on growth inhibition and gene expression in gastric carcinoma cell lines. Methods: Growth inhibition, cell malignant degree and gene expression were studied using MTT assay, colony formation in soft agar and Northern blot methods. Results: All trans retinoic acid (ATRA) effectively inhibited the growth of MGc80 3 and BGC 823 gastric carcinoma cells, whereas it had little effect on MKN45 gastric carcinoma cell. ATRA also decreased the colony formation ability of three gastric carcinoma cell lines. In MGc80 3 and BGC 823 cells, p53 expression level (wild type) was increased, and c myc expression level was decreased after induced by ATRA. In contrast, in MKN45 cell, p53 expression and c myc expression level was not affected by ATRA. Retinoic acid receptor α (RARα) expression level was low in three gastric carcinoma cell lines, it was increased obviously in MGc80 3 and BGC 823 cells, but not in MKN45 cell after induction of ATRA. Conclusion: The high level of RAR α expression induced by ATRA may correlate with growth inhibition and regulation of p53 and c myc expression by ATRA in gastric carcinoma cell lines.
