Preliminary studies of arsenic trioxide on the proliferationof neuroblastoma cells

Objective: To investigate the effect of arsenic trioxide(As2O3) on the proliferation of neuroblastoma (NB) cells and its mechanisms of action. THZMethods: 1The effect of As2O3 on the proliferation of NB cells were measured with MTT method. 2Morphological changes of the experimental and control groups were observed under the optical microscope in MayGrundwalGiemsa stain.3Ratio of apoptotic cell was determined by the flow cytometry assay in propidium iodide staining.4DNA fragmentation was observed on the gel electrophoresis. Results: 1As2O3 could significantly inhibit the proliferation of NB cell line SJNSH and the inhibitory effect was doseandtimedependent.2All results of morphology,the flow cytometry assay and the gel electrophoresis indicated that As2O3 induce apoptosis of SJNSH cells.Besides, that As2O3induced apoptosis of SJNSH cells was doseandtimedependent. Conclusion: As2O3 significantly inhibited the proliferation of SJNSH cells through inducing apoptosis in these cells.
