A method for determination of L-theanine with HPLC-PDAD in tea

Using trifluoroacetic acid and acetonitrile as the mobile phase,underivatized L-theanine in six categories of tea extracts were analyzed by HPLC-PDAD.The results showed that the UV detection wavelength of L-theanine was in the range of 190~240 nm with the maximum absorbance at 199 nm,and the calibration curve between peak area and contents was linear in the range of 0.02~1.00 mg·mL~(-1).The linear regression equation was Y=5.8343×10~6X+1.8201×10~5(r~2= 0.9991).RSD of L-theanine in each sample was 0.24%~1.74%,and the mean recovery rate was 99.21%~101.28%.
