Establishment of AFLP technique system for Dangshansu pear and its variants determination

The AFLP technique for pear genomic DNA analysis was established according to systematical study of the main factors involved in technique system.The digestion of genomic DNA was implemented by using two-step digestion with two restriction enzymes.500 ng DNA was digested with 5 units of EcoRⅠ and MseⅠ respectively in a reaction volume of 25 μL.Double-stranded adaptors were legated to the restriction fragments at 16℃ overnight(or 8~12 h).The digested-legated DNA fragment was diluted to 10 times with ddH_2O and used in pre-amplification.The pre-amplification products were diluted to 15 times with ddH_2O and used in selective amplification.The eight variants all have special bands in comparison with Dangshansu pear indicated that altered in genetics.The AFLP technique could be used to determine the mutants of pear trees.
