Callus Induction and Plantlet Regeneration of Eucalyptus urophylla×E.grandis through Tissue Culture

Stem sections and leaf discs of hybrid of E.urophylla×E.grandis were used as explants to induce callus and regenerate explants.The results indicated that callus could be induced from both of the explants in the improved H media in 7-10 days,the induction rates of the callus were over 90%. In the improved H media with 1.0 mg·L-1 16-BA, 0.5mg·L-1 NAA and 5% sugar, the adventitious buds from the young stem callus were induced and its differentiation rate was 22.2%. Each callus germinated 10-20 greenish shoots. Meanwhile the embryo-derived plantlets were obtained from the same stem callus, and the plantlets were formed with simultaneous development of roots and shoots.
