Protective effect of gypenosides on global cerebral ischemia-reperfusion injury in rat

Acute cerebral ischemia-reperfusion injury was produced in rat by clipping basilar artery and bilateral commoncarotid arteries for 60 minutes,and followed by removing the clips for another 60 minutes.Gypenosides in dosage of l00mg/kg body weight was injected intraperitoneally l5 minutes before clipping.Electroencephalography(EEG)wasmonitored,and cerebral water,Na+ and K+ content,lactate dehydrogenase(LDH)and creatine kinase(CK)activity,lactate and malondialdehyde(MDA)were measured to examine the protective mechanism of gypenosides on global cerebralischemia- reperfusion injury.The results showed that cerebral water and Na+ content were significantly lower ingypenosides group than in reperfusion group(P0.01,P0.01),lactate dehydrogenase and CK release from brainduring cerebral reperfusion were depressed significantly(P0.01),(P0.01),lactate and MDA level were falling ingypenoside(P0.0l,P0.01).These results indicate that gypenosides has a proctective effect on global cerebralischemia-reperfusion injury Its mechanism is related to the inhibition of brain edema,intracellular enzymes release,lactaeaccumulation and lipo-peroxidation.
