ESTABLISHMENT OF MODEL IN DIABETES MELLITUS COMPLICATED WITH INFECTION AND CHANGES OF TUMOR NECROSIS FACTOR α IN RATS

Objective To establish a rat model of diabetes mellitus(DM) complicated with infection, detect the changes of tumor necrosis factor α(TNFα) mRNA and protein in pancreas, and assess the inflammatory changes in this condition. Methods The rat model with diabetes mellitus complicated with infection was obtained by STZ-injection and CLP-operation. The rats were divided into normal, sham-operated, DM, infection, and DM complicated with infection(DMI) groups. The C-reactive protein(CRP), insulin and TNFα were measured by ELISA; semi-quantitative TNFα mRNA in pancreas was detected by RT-PCR. Results The rat models were established successfully. The levels of TNFα mRNA in pancreas and TNFα of plasma were higher than those in the normal, sham-operated and DM groups (H=7.56,q=3.25-5.26, P0.05;F=9.49, q=3.76-12.92,P0.05, 0.01),but not significantly different with those in the infection group (P0.05). The concentration of TNFα in the pancreas increased significantly in the DMI group (F=7.78,q=2.80-4.32,P0.05). Conclusion DM complicated with infection extended the inflammation reaction. The level of TNFαmay reflect the pancreatic injuries.
