CLINICAL SIGNIFICANCE OF MEASUREMENT OF MICROSATELLITE INSTABILITY OF TEL GENE IN CHILDHOOD ACUTE LYMPHOBLASTIC LEUKEMIA

ObjectiveTo investigate the correlation between TEL gene and the occurrence,progression and prognosis acute lymphoblastic leukemia(ALL) in children.MethodsMSI and LOH in 53 children with ALL,15 with benign hematopathies and five normal children were detected by multiplex-PCR amplification at four microsatellites linked to TEL gene on chromosome 12,gel electrophoresis and argentine dye.ResultsLOH in 23 cases(23/53),the frequency of LOH on four microsatellites(D12s89,D12s98,D12s269,D12s358) was 43.40%,among those microsatellites,the expression ratio of LOH on D12s89 was the highest(39.62%).Two cases were detected MSI on D12s89,D12s98,D12s269,its frequency was 3.77%.At the same time,LOH and MSI were dynamic observed in the three stages(preliminary,remission,relapse)in 18 cases of all the patients,the frequency of LOH in the three stages was 61.11%,11.11% and 66.67%,respectively(χ~2=8.88,11.69;P0.01).LOH and MSI were not found in preliminary stage,but LOH was found simultaneously in three loci in the relapse stage.LOH was consistently detected in the three stages in two cases.LOH and MSI were not found on the four sites in 15 cases of benign hematopathies and five healthy children.ConclusionTEL gene is related to the occurrence and development of childhood ALL.The detection of MSI can be used in the diagnosis,differential diagnosis,and prediction of the prognosis of the disease,which can also be used to detect minimal residual leukemia.
