Electrophoresis of bare and proteins-capped gold colloidal nanoparticles

Several sizes of gold colloidal nanoparticles ranging between5nm and50nm in dia-meter were prepared by using citrate reduction method.Each of them was capped with different proteins,respectively.Elec trophoresis of bar e and proteins-capped gold colloidal nanoparticles through agarose-gel support was carried out.At the same time ,AgNO 3 as well as hydroquinone served as signal enhancer.The samples were well dis played on the plate of agarose-gel,and their position distribution in gel tunn els were also clearly observed.The experimental results showed that,under an a pplied electric field,the smaller size of gold colloidal nanoparticles were,th e faster particles migrated.They were all presented dispersive distribution in agarose-gel.However,the same size of gold colloidal nanoparticles capped wit h proteins were well separated,and their electrophoretic behaviors were analogu es to the characteristic of protein moleculars,but not com-pletely identical with them.Among these particles,the migration speed of which capped with Bovin e serum albumin(BSA)was the fastest,and the order of other capped components was staphylococcal protein A(SPA),recombinant human calmodulin(rhCaM),as well as GAHIgG-? in turn.Furthermore,no dispersion phenomenon occurred.This result provide d a novel idea for separation of biological components,detection of partner mol ecules and preparation of micro/nano structures.
