STUDIES ON DUCK PLAGUE VIRUS DNA WITH RESTRICTION ENDONUCLEASES

Endonuclease maps of an attenuated duck plague virus ( DPV ) were reported.DPV was replicated in CEF culture.The harvested virus was concentrated by differential precipitation with ammonium sulphate and purified through 25-45%(w/v ) sodium potassium tartrate gradiants.The DPV DNA was isolated by means of protease digestion-SDS-phenol method before cleavage with restriction endonuclease EcoR Ⅰ,BamHI,Hind Ⅲ and Pst Ⅰ respectively. The DNA fragments ware resolved by 0.7% agarose gel electrophoresis. The molecular weight of DPV genome, estimated by summation of fragment sizes from cleavage patterns, ranged from 91.84×106 to 94.84×108. In addition, by isopycnic CsCl gradient centrifugation, the buoyant density of DPV particles also was determined as 1.272g/ml. The classification of DPV was discussed.
