SEQUENCE ANALYSIS OF HYPHANTRIA CUNEA NUCLEAR POLYHEDROSIS VIRUS CHITINASE GENE

Hyphantria cunea nuclear polyhedrosis virus chitinase A(ChiA) gene was located within a HindⅢ -3.5kb fragment. The DNA sequencing indicated that the ChiA gene was detected at the upstream of 5'-terminus of cysteine protease gene in the opposite orientation. The ChiA ORF of 1662nt encoded the protein of 553 amino acids. The TAAG motif and poly(A +) signal sequence AATAAA was found at 5' and 3' noncoding region of ChiA gene respectively. HcNPV ChiA gene showed 75.3%, 75.7% nucleotide homology and 75%, 78.7% amino acid homology respectively to those of Autographa californica multicapsid nuclear polyhedrosis virus and Bombyx mori nuclear polyhedrosis virus, and also showed 51% identity to bacterium Serratia marcescens ChiA at the amino acid level. It is possible that baculovirus chitinase gene and bacterial chitinase gene share a common ancestor.
