The Cytoplasmic Affinity of 38kD Phosphorylated Protein Gene Product of Marek's Disease Virus

To express Marek's disease virus(MDV) 38kD phosphorylated protein(pp38) gene by its own promoter,a recombinant expression plasmid pcDNA pp38 was constructed by inserting the whole pp38 gene fragment with its own upstream regulating sequence into BglⅡ and HindⅢ sites of pcDNA3.1/Zeo in which the pCMV fragment was deleted by BglⅡ and HindⅢ digestion.The transfection experiment indicated that the naive promoter of pp38 gene still worked well in CEF when it was inserted into the plasmid.The indirect fluorescence antiboldy test(IFA) with pp38 specific monoclonal antibody H19 demonstrated that the expressed pp38 mainly localized in the cytoplasm but not in the nucleus of the transfected CEF.It is in contrast to another MDV gene product meq,which mainly localized in the nucleus of the transfected CEF.The different locations of expressed meq and pp38 gene products represented their different biological functions.
