Study on Isolation and Identification of SARS Coronavirus in Experimentally Infected Rhesus Monkeys

We established methods for isolation and culturing of SARS-coronavirus(SARS-CoV) to supply experimental support for SARS animal model establishment and, also, the testing criteria were established based on survival time of SARS-CoV in vivo. The Rhesus monkeys were infected by inoculation via nasal cavity with identified SARS-CoV from SARS patient.Pharyngeal swabs and serum or plasma were collected in indicated times for virus isolation on Vero cells.The results showed that SARS-CoV were isolated from pharyngeal swabs at time points of 2,5,7 days after virus infection; from lung,spleen,liver,kidney and lymph node on 5~15 days after virus infection.The isolated virus was identified to be SARS-CoV by immunofluorescence assay and RT-PCR. We verified for the first time that SARS-CoV could replicate in Rhesus monkeys by these experiments.The successful isolation of SARS-CoV in Rhesus monkey is one of the chief supports for establishing a SARS-CoV-infected animal model, and also provides reliable criteria for evaluation on safety and validity of vaccines and drugs.
