EAG responses of the male cotton bollworm (Helicoverpa armigera) and the male oriental tobacco budworm (H.assulta) to female sex pheromones

Comparison of electroantennogram (EAG) responses between Helicoverpa armigera and H.assulta to female sex pheromone major components, (Z) 11 hexadecenal(Z 11 16:Ald) and (Z) 9 hexadecenal(Z 9 16:Ald) was investigated. The basal end of the isolated antennae was connected to the indifferent electrode, and the tip after the terminal segment removed was linked with the recording electrode. The antennae were stimulated by six dosages of sex pheromone components from 1 ng/μl to 10 5 ng/μl through a Pasteur pipette. The EAG signal was amplified by DC/AC amplifier and recorded by Spike software on a PC computer. The absolute EAG data were used for statistical analysis. The dose response curve showed that both H. armigera and H.assulta had high response amplitudes to Z 11 16: Ald, but that of H. armigera was higher than that of H. assulta from 10 2 ng/μl to 10 5 ng/μl. High spike amplitude was elicited in H. assulta by Z 9 16: Ald, but relative low response to Z 9 16: Ald in H. armigera was detected, this indicates that there is no Z 9 16:Ald sensillum on the H.armigera antennae. Both H. armigera and H. assulta had strong responses to mixtures of Z 11 16:Ald and Z 9 16:Ald with ratios 97∶3 and 7∶93, but with no significant difference between the two species. Results suggested that the EAG response of H. armigera and H. assulta could detect the single specific component of sex pheromones, but could not tell the difference of species specific pheromone blends.
